Expert View in ONdrugDelivery: Simulation-Led Development of Dual-Chamber Injection Systems

Veryst’s Dr. Matthew Hancock and Dr. Sahab Babaee of Sanofi have co-authored an ONdrugDelivery Expert View on simulation-led development of dual-chamber autoinjectors. Its full title is “Enabling Delivery of Lyophilised Biologics via Simulation-Led Development of Dual-Chamber Systems.”

Dual-chamber autoinjectors combine reconstitution and injection of lyophilized drugs in one patient-triggered device. This simplicity for the patient relies on a precise sequence of spring, stopper, air, and fluid events. Many failure modes only surface late in development.

The authors describe a validated reduced-order modeling framework for these devices. It predicts stopper motion, reconstitution and injection times, and tissue backpressure. Developers can explore design and formulation tolerances computationally. They can then narrow the design space before committing to tooling or physical testing. The article also features Veryst’s Dual Chamber Cartridge Simulator, a browser-based version of the model.

They show how this approach aligns with ASME V&V 40 and FDA credibility guidance. Finally, they outline future opportunities in high-concentration biologics, mRNA vaccines, and connected autoinjectors.
